Nagarjuna was born on 29 August 1959 in Chennai, Tamil Nadu, India. He is the son of the late actor Akkineni Nageswara Rao and his wife Annapurna Akkineni. He has two brothers, Venkat and Sumanth, and a sister, Supriya. He attended the Little Flower Junior College in Hyderabad and graduated with a degree in commerce from the Nizam College in Hyderabad.

Nagarjuna made his acting debut in 1986 with the Telugu film Vikram. He went on to star in several successful films, including Geetanjali (1989), Shiva (1990), and Ninne Pelladata (1996). He also starred in the critically acclaimed films Annamayya (1997) and Manam (2014). He has also produced several films, including Annamayya (1997), Manam (2014), and Soggade Chinni Nayana (2016).
